阪崎腸桿菌CDC3225-73ATCC菌種（又稱(chēng)阪崎氏腸桿菌）是腸桿菌科的一種，1980年由黃色陰溝腸桿菌更名為阪崎腸桿菌。阪崎腸桿菌能引起嚴(yán)重的新生兒腦膜炎、小腸結(jié)腸炎和菌血癥，死亡率高達(dá)50% 以上。目前，微生物學(xué)家尚不清楚阪崎腸桿菌的污染來(lái)源，但許多病例報(bào)告表明嬰兒配方粉是目前發(fā)現(xiàn)的主要感染渠道。
阪崎腸桿菌的檢驗(yàn)范圍：本標(biāo)準(zhǔn)規(guī)定了食品中 阪崎腸桿菌（Enterobacter sakazakii）的檢驗(yàn)方法，適用于嬰幼兒配方食品、乳和乳制品及其原料中阪崎腸桿菌的檢驗(yàn)。
一、設(shè)備和材料
除微生物實(shí)驗(yàn)室常規(guī)滅菌及培養(yǎng)設(shè)備外，其他設(shè)備和材料如下：
1 冰箱：2 ℃～5 ℃。
2 恒溫水浴箱：44 ℃±0.5 ℃。
3 恒溫培養(yǎng)箱：25 ℃±1 ℃，36 ℃±1 ℃，44 ℃±0.5 ℃
4 天平：感量0.1 g。
5 均質(zhì)器。
6 振蕩器。
7 無(wú)菌吸管：1 mL（具0.01 mL 刻度）、10 mL（具0.1 mL 刻度）或微量移液器及吸頭。
8 無(wú)菌錐形瓶：容量100 mL、200 mL、2000 mL。
9 無(wú)菌培養(yǎng)皿：直徑90 mm。
10 pH 計(jì)或pH 比色管或精密pH 試紙。
11 全自動(dòng)微生物生化鑒定系統(tǒng)。
二、培養(yǎng)基和試劑
1 緩沖蛋白胨水（buffer peptone water，BPW）
2 改良月桂基硫酸鹽胰蛋白胨肉湯-萬(wàn)古霉素（modified lauryl sulfate tryptose broth-vancomycin
medium，mLST-Vm）
3 阪崎腸桿菌顯色培養(yǎng)基。
4 胰蛋白胨大豆瓊脂（trypticase soy agar，TSA）
5 氧化酶試劑
6 L-鳥(niǎo)氨酸脫羧酶培養(yǎng)基：
7 糖類(lèi)發(fā)酵培養(yǎng)基：
8 西蒙氏檸檬酸鹽培養(yǎng)基
9 L-賴(lài)氨酸脫羧酶培養(yǎng)基：
10 生化鑒定試劑盒。
11 L-精氨酸雙水解酶培養(yǎng)基
三、檢驗(yàn)程序及操作步驟

2、 操作步驟
a. 前增菌和增菌
取檢樣100 g（mL）加入已預(yù)熱至44 ℃裝有900 mL 緩沖蛋白胨水的錐形瓶中，用手緩緩地?fù)u動(dòng)至充分溶解，36 ℃±1 ℃培養(yǎng)18 h±2 h。移取1 mL 轉(zhuǎn)種于10 mL mLST-Vm 肉湯，44 ℃±0.5 ℃培養(yǎng)24 h±2h。
b.分離
c. 輕輕混勻mLST-Vm 肉湯培養(yǎng)物，各取增菌培養(yǎng)物1 環(huán)，分別劃線(xiàn)接種于兩個(gè)阪崎腸桿菌顯色培養(yǎng)基平板，36 ℃±1 ℃培養(yǎng)24 h±2 h。
d. 挑取1 個(gè)～5 個(gè)可疑菌落，劃線(xiàn)接種于TSA 平板。25 ℃±1 ℃培養(yǎng)48 h±4 h。
e.鑒定
自TSA 平板上直接挑取黃色可疑菌落，進(jìn)行生化鑒定。阪崎腸桿菌的主要生化特征見(jiàn)表1?？蛇x擇生化鑒定試劑盒或全自動(dòng)微生物生化鑒定系統(tǒng)。
