ATCC medium: 1210 Avian mycoplasma medium
PPLO Broth w/o CV (BD 255420)...................................14.7 g
Distilled or deionized water...................................650.0 ml
Swine or horse serum (heat-inactivated)........................150.0 ml
Yeast Extract Solution (GIBCO 18180)...........................100.0 ml
Glucose solution [10% (wt/vol)].................................10.0 ml
Arginine solution [10% (wt/vol)]................................10.0 ml
NAD [1% (wt/vol)]...............................................10.0 ml
Phenol red solution [0.1% (wt/vol)] (for liquid medium only)....20.0 ml
Agar (for solid medium only)*...................................10.0 g
Mix the PPLO broth base in the water (together with agar for solid
medium) and autoclave at 121C for 15 minutes. Mix the remaining
ingredients and heat to 56C in a water bath. Combine both lots of
ingredients, mix, and dispense.
*Use agar known to be noninhibiting to mycoplasmas.
