Solution A:

Na2S2O3 . 5H2O ......................5.0 g

NH4Cl ..............................1.0 g

KNO3 ...............................2.0 g

Distilled water..................250.0 ml

Solution B:

KH2PO4 .............................2.0 g

Distilled water..................250.0 ml

Solution C:

NaHCO3 .............................2.0 g

Distilled water..................250.0 ml

Solution D:

MgSO4 . 7H2O .......................0.8 g

FeSO4 . 7H2O (2%, w/v, in N HCl) ...1.0 ml

Trace Metals Solution (see below)..1.0 ml

Distilled water..................250.0 ml

Sterilize the four solutions separately and combine aseptically for the completed medium.

The pH of the final medium is 7.0.

Trace Metal Solution:

EDTA..............................50.0 g

ZnSO4 .............................22.0 g

CaCl2 ..............................5.54 g

MnCl2 ..............................5.06 g

FeSO4 ..............................4.99 g

Ammonium molybdate.................1.10 g

CuSO4 ..............................1.57 g

CoCl2 ..............................1.61 g

Distilled water....................1.0 L

Adjust pH to 6.0 with KOH.
