Comments:The NC-37 line has been utilized as a target cell for EBV superinfection studies and has proven useful in chemical induction studies of the latent EBV genome and in the propagation of various (retroviruses) oncornaviruses. The consistency of the line's characteristics has made it a useful tool in a wide variety of biochemical, immunological, virological and cell biological studies.
Each cell contains an average of 60 EBV genome copies. The cells are EBNA positive (EBNA+) and surface immunoglobulin negative (sIg-).
Although deposited with the ATCC as a lymphoblastoid cell line established from a 34 year old male caucasian, DNA fingerprinting has shown this line to be a derivative of Raji (see ATCC CCL-86 ), a Burkitt's lymphoma cell line.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Temperature: 37.0°C
Subculturing: Protocol: Cultures can be maintained by addition or replacement of fresh medium. Maintain cultures at 5 X 10(5) viable cells/ml. A maximum density of upto 2 X 10(6) is possible.
Medium Renewal: Add fresh medium (20% to 30% by volume) every 3 to 4 days
Preservation: Complete growth medium supplemented with 5% (v/v) DMSO.
