Comments:The line was isolated from a patient who was negative for anti human T lymphotropic virus type I (HTLV-I) antibody, and no HTLV-I sequences were detected in the cultured cells by Southern analysis. Although HH cells lack the alpha chain of the IL-2 receptor (IL-2R alpha), they do possess a functional p70 beta chain (IL-2R beta) of the IL-2 receptor complex.
A culture submitted to the ATCC in June of 1992 was found to be contaminated with mycoplasma, and was cured by treatment with Ciprofloxacin.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Atmosphere: air, 95%; carbon dioxide (CO2), 5%
Temperature: 37.0°C
Subculturing: Protocol: Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 10(5) cells/ml and maintain between 1 X 10(5) and 1 X 10(6) cells/ml.
Medium Renewal: Every 2 to 3 days
Preservation: Freeze medium: Complete growth medium, 95%; DMSO, 5%
Storage temperature: liquid nitrogen vapor phase
