Isolation: Isolation date: 1984
Comments:Animals were immunized with purified exotoxin A of Pseudomonas aeruginosa.
Spleen cells were fused with Sp2/0-Ag14 cells.
The antibody binds specifically for the exotoxin A of Pseudomonas aeruginosa.
It displays specificity for SDS-denatured exotoxin A from the strain PA103 and its derivative PA103/29 but does not react with exotoxin from strain PAO1 or its derivative PAO-PR1. [40007 ]
The antibody shows high specificity and reactivity for denatured exotoxin A in immunoblotting and ELISA assays.
Both TC1 (ATCC CRL-2493 ) and TC31 (ATCC CRL-2494 ) antibodies neutralize exotoxin A toxicity against CHO cells in an in vitro assay but only TC31 protected mice infected with strain PA220 in an in vivo model. [40007 ]
Propagation: ATCC complete growth medium: Iscove's modified Dulbecco's medium with 4 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 85%; fetal bovine serum, 15%

Subculturing: Protocol: Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 5 X 10(4) viable cells/ml. Do not allow the cell concentration to exceed 5 X 10(5) viable cells/ml.
Interval: Maintain cultures at a cell concentration between 5 x 10(4) and 5 X 10(5) cells/ml.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
Preservation: Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
