Comments:Animals were immunized with human tonsillar lymphocytes.
Spleen cells were fused with NS-1 myeloma cells.
The antibody is specific for the Bp50 (CD40) B cell surface antigen.
The antibody reacts specifically with CD20 positive (Bp35, B1) blood and tonsillar B cells, and does not react with T cells or NK cells.
It does react with malignant B cells and B cell lines.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Temperature: 37.0°C
Subculturing: Protocol: Cultures can be maintained by the addition of fresh medium or by replacement of medium. Alternatively, cultures can be established by centrifigugation with subsequent resuspension at 2 X 10(5) viable cells/ml. Maintain between 1 X 10(5) and 1 X 10(6) viable cells/ml.
Medium Renewal: Every 2 to 3 days
Preservation: Freeze medium: culture medium, 92.5%; DMSO, 7.5%
