Comments:TGP55 is an epithelial-like cell line derived from a pancreatic tumor with small cell anaplastic morphology that arose in an adult female transgenic mouse.
The mouse carried the pseudogene construct composed of elastase-1 promoter linked to the coding region for the SV40 T antigen.
The cells do not become confluent.
They grow very slowly as islands, and should be subcultured when the islands begin to form domes.
Propagation: ATCC complete growth medium: A 1:1 mixture of Dulbecco's modified Eagle's medium and Ham's F12 medium, 90%; heat-inactivated fetal bovine serum, 10%
Subculturing: Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:4 is recommended
Medium Renewal: Twice per week
Remove spent medium, add fresh 0.25% trypsin, 0.03% EDTA solution, rinse and remove trypsin. Let the culture sit at room temperature (or at 37C) until the cells detach. Add fresh medium, aspirate and dispense into new flasks.
Preservation: Culture medium, 95%; DMSO, 5%
