Comments:GC-2spd(ts) was established by stable cotransfection of freshly isolated spermatocytes with the SV40 large T antigen gene (pSV3neo, see ATCC 37150 ) and a temperature sensitive mutant of the p53 tumor suppressor gene (LTRp53cG9).
Cells were selected with G-418, cultivated for 6 months and single cell cloned three times by limiting dilution.
No clonal proliferation was observed in soft agar cultures, indicating that these cells were immortalized but not transformed.
The cells have lost their differentiation potential, and are currently arrested at a premeiotic stage.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Temperature: 37.0°C
Subculturing: Protocol: Remove medium, and rinse with 0.25% trypsin, 0.03% EDTA solution. Remove the solution and add an additional 1 to 2 ml of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37C) until the cells detach. Add fresh culture medium, aspirate and dispense into new culture flasks.
Subcultivation Ratio: A subcultivation ratio of 1:5 to 1:7 is recommended
Medium Renewal: Every 2 to 3 days
Preservation: Freeze medium: Complete growth medium, 95%; DMSO, 5%
