Spleen cells were fused with P3X63Ag8.653 myeloma cells.
The antibody reacts with rboIL-1 beta and cross-reacts with its human analog (rHuIL-1 beta).
It does not react with other related monokines tested (ie.g., human IL-6 and TNF alpha).
The antibodies neutralizes IL-1 activity (both macrophage derived IL-1 and recombinant IL-1 beta) in a thymocyte costimulation assay.
Propagation: ATCC complete growth medium: Dulbecco's modified Eagle's medium with 4.5 g/L glucose, 1 mM non-essential amino acids, 0.1 mM hypoxanthine, 0.032 mM thymidine and BME vitamins, 70%; hybridoma cloning factor (Origen), 10%; fetal bovine serum, 20%
Subculturing: Medium Renewal: Every 2 to 3 days
Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 10 exp5 cells/ml and maintain between 1 X 10 exp5 and 1 X 10 exp6 cells/ml.
