Spleen cells were fused with P3X63Ag8.653 myeloma cells.
The antibody reacts with B6 vitamers.
It binds to both pyridoxine and pyridoxamine with similar affinities.
Since the product binds an essential nutrient, the medium must be changed frequently or the cells will die.
Propagation: ATCC complete growth medium: Dulbecco's modified Eagle's medium with 4.5 g/L glucose, and 0.1 mM non-essential amino acids, 70%; NCTC 109 medium, 10%; fetal bovine serum, 20%
Subculturing: Medium Renewal: 3 times per week
Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 1 X 10 exp5 cells/ml and maintain between 1 X 10 exp5 and 1 X 10 exp6 cells/ml.
Preservation: fetal bovine serum, 95%; DMSO, 5%
