Comments:Animals were immunized with a synthetic peptide with the sequence YTLVREIRQYRLKKESKEEKTPGC corresponding to peptides 157-180 of the v-ras K oncogene.
Spleen cells were fused with Sp2/0 myeloma cells.
The antibody reacts with the synthetic v-ras K oncogene peptide. In ELISA testing, the antibody weakly detects the homologous region in the ras H sequence. In immunoblot testing a serum protein of approximately 45 kd is detected.
The IgG1 and IgG2b reactivities of the antibody with the v-ras K peptide (157-180) was confirmed by ELISA. IgG1 was strongly positive while IgG2b was only slightly positive.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Temperature: 37.0°C
Subculturing: Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 X 10 exp5 viable cells/ml.
Maintain cell density between 2 X 10 exp5 and 2 X 10 exp6 viable cells/ml.
Preservation: culture medium 95%; DMSO, 5%
