Comments:Animals were immunized with immunopurified human CR3 [PubMed: 1371129]. Spleen cells were fused with Sp2/0 myeloma cells. KIM127 stimulates homotypic aggregation by a CD11 pathway in the human B lymphoblastoid cell line, JY. The antibody recognizes an epitope on the Leukocytic cell-adhesion molecule common beta-chain (CD18) and promotes both lymphocyte function-associated antigen (LFA-1) and CR3-dependent adhesion events. The antibody recognizes a temperature-sensitive epitope that is not dependent on the presence of divalent cations [PubMed: 1371129]. The antibody recognizes an epitope distinct from the KIM185 (ATCC CRL-2839 ) antibody.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Atmosphere: air, 95%; carbon dioxide (CO2), 5%
Temperature: 37.0°C
Subculturing: Protocol: Shake off attached cells and scrape if necessary.Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 3 X 10(4) to 5 X 10(4) viable cells/ml.
Interval: Maintain cultures at a cell concentration between 8 X 10(4) and 5 X 10(5) cells/ml.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
