Comments:Animals were immunized with a pool of plasmacytoma cells (U266, RPMI1860 and OPM) in the left footpad and with K562 chronic myelogenous leukemia cells in the right footpad. The immunization was repeated after 14 days. Lymph node cells were fused Sp2/0 mouse myeloma cells. The antibody is reactive with cell surface membranes of both human myeloma cells and ovarian cancer cells. MA69 detects a distinct band on myeloma membranes binding to a cell surface glycoprotein with an estimated molecular weight of between 78 and 120 kDa. The antibody also recognizes an antigen on ovarian cancer cells with a molecular weight of 76 kDa to 213 kDa. The antigen shares at least one epitope with the multiple myeloma surface antigen. MA69 is not reactive with cell surface membranes of human peripheral blood mononuclear cells, human B cells, neuroblastoma cells, human B cell myelogenic leukemia cells, breast cancer cells, prostate cancer cells, or cervical cancer cells.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Atmosphere: air, 95%; carbon dioxide (CO2), 5%
Temperature: 37.0°C
Subculturing: Protocol: Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 5 X 10(4) viable cells/ml. Adherent cells may be removed by shaking the flask.
Interval: Maintain cell density between 5 X 10(4) and 5 X 10(5) viable cells/ml.
Medium Renewal: Every 2 to 3 days
Preservation: Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
Storage temperature: liquid nitrogen vapor phase
