Comments:The cells have a high level of oxidative activity, and and can be used in mutagenesis assays for substances that are metabolized into mutagenic products. They are are sensitive to killing by benzo[a]pyrene. The cells carry an endogenous Epstein-Barr virus (EBV) genome and are EBNA positive.
Propagation: ATCC complete growth medium: RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, and 1.0 mM sodium pyruvate, 90%; horse serum, 10%
Atmosphere: air, 95%; carbon dioxide (CO2), 5%
Growth Conditions: The cells grow as floating aggregates of large cell clusters.
Subculturing: Protocol: Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 to 2 X 10(5) viable cells/ml. Maintain cell density between 1 X 10(5) and 1 X 10(6) viable cells/ml.
