Comments:HCC1954 BL is a B lymphoblastoid cell line initiated from peripheral blood lymphocytes from the same patient as HCC1954 (ductal carcinoma of the breast) by transformation with Epstein-Barr virus (EBV).
The cell line was derived from a 61 year-old East Indian female with stage IIA, grade 3, primary ductal carcinoma of the breast.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Temperature: 37.0°C
Subculturing: Protocol: Cultures can be maintained by addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension in fresh medium at 3 to 5 x 10 exp5 viable cells/ml. Maintain cultures at cell concentrations between 5 X 10 exp5 and 2 X 10 exp6 viable cells/ml.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density)
Preservation: Freeze medium: Complete growth medium, 95%; DMSO, 5%
Storage temperature: liquid nitrogen vapor phase
