Glucose............................5.0 g
Apple juice......................200.0 ml
5% Tween 80........................1.0 ml
Agar (see comments below).........20.0 g
Distilled water..................800.0 ml
Dissolve liver extract in approximately 100 ml distilled water with stirring. Remove any undissolved material by filtration through Whatman #1 paper. Bring volume to 800 ml with additional distilled water. Add remaining ingredients. Adjust to pH 4.5 with H3PO4. Autoclave at 121C for 15 minutes. Comments: To prepare solid medium, dissolve agar with heat and stirring in 500 ml distilled water. Prepare remaining ingredients in a total volume of 500 ml solution.
Autoclave agar and nutrient solutions separately at 121C for 15 minutes. Cool to 50C. Aseptically combine the two solutions, mix well, and dispense.
