Heart Infusion Broth (BD 238400)........ 25.0 g
Hemoglobin (BD 213640)................... 2.0 g
Mucin Bacteriological (BD 212111)........ 5.0 g
Distilled water.......................... 1.0 L
Adjust pH to 7.8. Heat mixture at 93-95C for 30 minutes in a water bath. Restore to original volume. Add 0.5% Celite (Analytical Filteraid, Johns Manville). Mix thoroughly. Filter through Whatman GFA (glass fiber paper) in Buchner filter. Clarify using 0.45 microns Millipore filter. Add 15% inactivated turkey serum. Filter-sterilize using positive pressure. For solid medium: Prepare 42.5 ml double strength broth and 42.5 ml distilled water containing 0.7 g Agar, Noble (BD 214230). Sterilize the solutions separately and combine aseptically at 56C with 15 ml sterile turkey serum for a final volume of 100 ml.
