ATCC medium: 359 Modified TYM basal medium (ATCC medium 358) with pH adjusted to 7.2 and 0.2-0.5 ml of heat-inactivated horse serum added per tube before use
ATCC medium 358 (see below); adjust pH to 7.2; add 0.2-0.5 ml of heatinactivated horse serum per tube before use.
ATCC Medium 358 Modified TYM Basal Medium:
Trypticase Peptone (BD 211921)..........20.0 g
Yeast extract...........................10.0 g
Maltose..................................5.0 g
L-Cysteine . HCl.........................1.0 g
L-Ascorbic acid..........................0.2 g
K2HPO4 ...................................0.8 g
KH2PO4 ...................................0.8 g
Agar, Noble (Difco 0142).................0.5 g
Distilled water........................900.0 ml
1. Add all the dry materials except the agar to the distilled water.
2. Adjust to the desired pH with 20% NaOH.
3. Add the agar and heat to dissolve.
4. Tube in 9.0 ml volumes in 16 X 125 mm screw-capped test tubes and autoclave for 15 minutes at 121C. Leave caps loose while autoclaving. Tighten the caps immediately upon removing the tubes from the autoclave. This medium can be stored for about one month under refrigeration. Cells grow poorly or not at all in old medium. Keep tubes tightly capped at all times.
