培養(yǎng)方法
挑取活化好的菌種接入種子培養(yǎng)基，30攝氏度，160r/min,振蕩培養(yǎng)24h后以6%的接種量接入液體培養(yǎng)基，30攝氏度下靜置培養(yǎng)7天。
樣品純化處理方法
將培養(yǎng)木醋桿菌所得凝膠膜經(jīng)蒸餾水漂洗，1 %氫氧化鈉溶液煮沸4h，于室溫下在0.5%乙酸中浸泡0.5H，蒸餾水沖洗烘干后稱重。
樣品的性能測試
細菌纖維素樣品用變換紅外光譜儀分析有機陰離子官能團，測試波數(shù)范圍為4000-400cm。用電子顯微鏡觀察樣品的表面形貌。熱重分析采用熱重分析儀,稱取2.031mg粉末，升溫速度10攝氏度/min，檢測溫度25-45攝氏度。
最佳培養(yǎng)環(huán)境
葡萄糖30g/L，蛋白胨7.5g/L，酵母粉10g/L，磷酸氫二鈉7.5g/L，檸檬酸0.5g/L，PH0.5。 五個因素對細菌纖維素產(chǎn)量的影響次序為：酵母粉>PH值>葡萄糖>檸檬酸>蛋白胨。
