Incubation : Temperature: 31.0°C Duration: 1-3 days; For best results, cells should be 24 to 48 hours old and 80-90% confluent at time of infection [not 100% confluent].
Comments : Selected after mutagenesis with nitrosoguanidine and replication in the presence of bromodeoxyuridine. The non-permissive incubation temperature is 39.5oC. This mutant was not assigned a Dales EM category. The mutation was assigned by complementation analysis (Lackner, et al, 2003) to vaccinia map location U10. Vaccination recommended for lab staff manipulating vaccinia, monkeypox, or cowpox viruses (CDC/NIH Biosafety in Microbiological and Biomedical Laboratories, 4th ed, 1999 - stock no. 017-040-00547-4).
