Comments : This conditional lethal mutant, originally designated plaque isolate no. 6389, was gnerated from the IHD-W strain of Vaccinia by S. Dales by mutagenesis with nitrosoguanidine and replication in the presence of bromodeoxyuridine. It was assigned to Dales EM category C, indicating that it induces rudimentary virus membranes with spicules in foci of viroplasm in infected cells. The mutation was assigned by complementation analysis (Lackner, et al, 2003) to vaccinia map location D5, an ATPase required for DNA replication. Shifting infection conditions from permissive to non-permissive temperature results in an abrupt cessation of DNA synthesis, described as a fast stop phenotype.Vaccination recommended for lab staff manipulating vaccinia, monkeypox, or cowpox viruses [70405]
