Host Organism : Production Host: MA104.1 (ATCCCRL-2378.1) Alternate Hosts: BS-C-1 (ATCCCCL-26), Rhesus embryonic monkey kidney cells, Primary cynomolgus monkey kidney cells
Incubation : For best results, plate cells 24-48 hours in advance. Infect at 80%-90% confluent, not 100% confluent. Remove medium and inoculate with a small volume of virus diluted to provide a MOI of about 0.1. Adsorb 1 hour at 37.0°C in a humidified 5% CO2 atmosphere, rocking every 20-30 minutes to redistribute inoculum. End adsorption by adding virus growth medium. Incubate at 37.0°C in a humidified, 5% CO2 atmosphere for 2-3 days, until CPE are well advanced in 100% of the culture. In TC, ATCCVR-1739 should be grown in serum free media that contains 2.0μg/ml trypsin.
Effect : CPE in the form of refractile, rounded cells throughout monolayer or in loose clusters.

Comments : Derived by adaptation of primary cell culture product ATCCVR-954 to MA104.1 (ATCCCRL-2378.1) cells. (Note: ATCCVR-954 and ATCCVR-1739 have not been compared with respect to sequence or infectivity in primary cell culture and tissue culture).
