Derived by adaptation of CHSE (ATCC® CRL-1681™) cell culture product ATCC® VR-1387™ to EPC cells (ATCC® CRL-2872™). ATCC® VR-1387™ and ATCC® VR-1872™ have not been compared for sequence or infectivity.
Host
EPC [Epithelioma Papulosum Cyprini] (ATCC CRL-2872)
Effects
cell degeneration; cell rounding; CPE
Complete medium
EMEM (ATCC® 30-2003™) + 5% FBS (ATCC® 30-2020™) + 20 mM HEPES (Sigma-Aldrich® 83264-100ML-F)
Temperature
15°C
Incubation
2-4 days at 15°C in a humidified atmosphere, until CPE is progressed through 80% of the monolayer.
Recommendation for infection
Plate cells 18-24 hours prior to infection and infect when cultures are 80-90% confluent. Remove medium and inoculate with a small volume of virus (e.g., 1 mL per 25 cm2) diluted to provide an optimal MOI (e.g., 0.01). Adsorb 1-2 hours at 15°C in a humidified atmosphere. End adsorption by adding virus growth medium.
